Target Information
Beta-arrestin-2 (ARRB2) is an adaptor protein of the arrestin family containing N- and C-arrestin domains. It plays a regulatory role in heterotrimeric G protein-coupled receptor desensitization and modulates beta2-adrenergic receptor (β2AR) function by enhancing β2AR-mediated nuclear translocation of ERK. ARRB2 binds phosphorylated β2ARs, reducing their ability to activate G(S) proteins. Together with AIP4, ARRB2 mediates endosomal sorting of CXCR4 into a degradative pathway and may participate in hormone-specific desensitization of TSH receptors. It is also a Ca²⁺-binding protein in retinal outer segments, interacting with P-rhodopsin and contributing to synaptic transmission in photoreceptor cells. ARRB2 is widely expressed, particularly in neuronal tissues and spleen.
