Steroid receptors are ligand-dependent intracellular proteins that stimulate transcription of specific genes by binding to specific DNA sequences following activation by the appropriate hormone. Prior to activation, steroid receptors associate with various proteins in both stable and transient fashion. These include heat shock proteins (HSP70, HSP90), immunophilins (FKBP52, FKBP54, CyP-40), and proteins termed p23, p60, and p48.

p23, along with HSP70, HSP90, and p60, forms a transient intermediate complex with the progesterone receptor (PR). The cloned human p23 encodes a 160 amino acid protein highly conserved across species, with no homology to previously identified proteins. It is a highly acidic phosphoprotein possessing an aspartic acid-rich C-terminal domain and multiple phosphorylation sites. In vitro studies indicate p23 binds to HSP90 and is required for HSP90 and CyP-40 binding to PR. Although its exact function remains unclear, p23 is considered an important factor in PR function.
