Product Specific Information

This antibody detects endogenous levels of RIPK2 only when phosphorylated at Ser531.

Target Information

RIPK2 is a serine/threonine kinase involved in cell activation, proliferation, differentiation, and apoptosis. Upon stimulation by bacterial peptidoglycans, NOD1 and NOD2 are activated, oligomerize, and recruit RIPK2 through CARD-CARD domains. RIPK2 contributes to tyrosine phosphorylation of ARHGEF2 through Src kinase, leading to NF-kappaB activation by NOD2. Once recruited, RIPK2 autophosphorylates and undergoes Lys-63-linked polyubiquitination by E3 ligases XIAP, BIRC2, and BIRC3. The polyubiquitinated RIPK2 mediates recruitment of MAP3K7/TAK1 to IKBKG/NEMO, inducing further Lys-63-linked polyubiquitination and activation of IKBKB/IKKB. Consequently, NF-kappaB is released, translocates to the nucleus, and activates transcription of genes involved in immune response, growth control, and apoptosis protection. RIPK2 also plays a role during T-cell receptor engagement by promoting BCL10 phosphorylation and subsequent NF-kappaB activation.
