Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It acts as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and mediating iron recycling through endocytosis and heme breakdown.

CD163 is expressed on the surface of human monocytes and macrophages, particularly in the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, except in mantle zones and germinal centers of lymphoid follicles, interdigitating reticulum cells, and Langerhans cells. Expression is high on CD14⁺ and CD16⁺ monocytes and is upregulated by IL-10, IL-6, and dexamethasone.

LPS and PMA induce shedding of CD163 into plasma or supernatant. Binding of hemoglobin/haptoglobin complexes is calcium- and pH-dependent, with higher affinity for multimeric haptoglobin (HP1F phenotype). CD163 activation triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
