Target Information
IL-1 receptor associated kinases (IRAKs) are serine/threonine kinases involved in signaling via Toll/IL-1 pathways. IRAK1 is rapidly recruited by MYD88 to the receptor-signaling complex upon TLR activation. Its association with MYD88 leads to phosphorylation by IRAK4 and subsequent autophosphorylation and kinase activation.
IRAK1 can also phosphorylate TIRAP to promote its ubiquitination and degradation, and phosphorylates interferon regulatory factor 7 (IRF7) to induce activation and nuclear translocation, resulting in transcriptional activation of type I IFN genes, driving antiviral responses.
