Target Information
IL-1 receptor associated kinases (IRAKs) are serine/threonine kinases involved in signaling via Toll/IL-1 pathways. IRAK1 is rapidly recruited by MYD88 to the receptor-signaling complex upon TLR activation. Its association with MYD88 leads to phosphorylation by IRAK4 and subsequent autophosphorylation and kinase activation. Additionally, IRAK1 phosphorylates TIRAP, promoting its ubiquitination and degradation. IRAK1 also phosphorylates interferon regulatory factor 7 (IRF7), inducing activation and nuclear translocation, resulting in transcriptional activation of type I IFN genes and antiviral response.
