Target Information
IRS-1 is a major substrate of the insulin receptor and becomes phosphorylated in response to insulin, IGF-1, and IL-4 stimulation. It is phosphorylated on serine, threonine, and tyrosine residues in various tissues. Casein kinase II mediates part of the insulin-stimulated serine/threonine phosphorylation of IRS-1 in vivo. Thr502 is identified as a major phosphorylation site, with Ser99 as an additional site. IRS-1 contains three putative 14-3-3 binding sites (Ser270, Ser374, Ser641), with the Ser270 motif located in the phosphotyrosine binding domain responsible for insulin receptor interaction.
