Product Specific Information

This antibody detects endogenous levels of Cyclin D1 only when phosphorylated at Serine 90.

Target Information

Cyclin D1 (PRAD-1, bcl-1) is a key regulator of the cell cycle, functioning with CDK4 and/or CDK6 to phosphorylate the Rb protein. It acts as a proto-oncogene and is overexpressed in many human neoplasms, including mantle cell lymphomas (MCL). Cyclin D1 positively regulates protein phosphorylation, mammary gland epithelial proliferation, and adipocyte differentiation.

The CCND1 gene encoding Cyclin D1 is located on chromosome 11. Cyclin D1 is present in both the cytoplasm and nucleus, synthesized during G1 phase, and forms complexes with CDK4 or CDK6 in response to growth factor stimulation. These complexes inactivate Rb through phosphorylation and modulate CDK inhibitors such as p21Cip1 and p27Kip1. Without growth factor signaling, Cyclin D1 undergoes ubiquitin-mediated degradation, leading to G1/S phase arrest.

Cyclin D1 belongs to the conserved cyclin family, characterized by periodic protein expression during the cell cycle. It serves as a regulatory subunit of CDK4/6, essential for G1/S transition. Mutations, amplification, or overexpression of Cyclin D1 are frequently observed in tumors and contribute to tumorigenesis. Cyclin D1 is a valuable tool for cell cycle and cancer biology research.
