Product Specific Information
The antibody was affinity-purified from rabbit antiserum by affinity chromatography using an epitope-specific immunogen. The purity is greater than 95% as determined by SDS-PAGE.
Target Information
The mammalian homologues of the key cell death gene CED-4 in C. elegans have been identified in human and mouse and designated Apaf-1 (apoptosis protease-activating factor 1). Apaf-1 binds to cytochrome c (Apaf-2) and Caspase-9 (Apaf-3), leading to Caspase-9 activation. Activated Caspase-9 subsequently cleaves and activates Caspase-3, a key protease responsible for the proteolytic cleavage of many proteins during apoptosis.
Recently, Cho et al. identified a new Apaf-1 interacting protein (APIP), also known as CG129 and MMRP19, which acts as a negative regulator of ischemic injury. APIP competes with the Caspase-9 binding site of Apaf-1. APIP is predicted to encode a 204 amino acid protein. An isoform, APIP2, encodes a 242 amino acid protein, differing in its N-terminus due to alternative splicing. APIP transcripts are ubiquitously expressed in most adult tissues, with high expression in skeletal muscle, heart, and kidney.
