Pregnane X Receptor (Steroid and Xenobiotic Receptor), Ligand Binding Domain (PXR (SXR)-LBD) is a 64.7 kDa recombinant human protein (amino acids 111–434) expressed as a GST fusion protein in baculovirus-infected insect cells.
PXR (SXR)-LBD was expressed and purified in the absence of exogenous ligands, making it ideal for ligand binding and coregulator recruitment or displacement studies.

Target Information

This gene product belongs to the nuclear receptor superfamily, members of which are transcription factors characterized by a ligand-binding domain and a DNA-binding domain.
The encoded protein regulates transcription of the cytochrome P450 gene CYP3A4, binding to its promoter response element as a heterodimer with the 9-cis retinoic acid receptor (RXR).
It is activated by compounds that induce CYP3A4, such as dexamethasone and rifampicin.
Several alternatively spliced transcripts encoding different isoforms—some using non-AUG initiation codons—have been described. Additional transcript variants exist but are not fully characterized.
