Target Information
Cas9은 세균의 CRISPR(Clustered Regularly Interspaced Short Palindromic Repeats) 적응 면역 시스템과 관련된 RNA 가이드 DNA 엔도뉴클레이스 효소입니다. RNA를 이용해 특정 DNA 서열을 표적화할 수 있어 유전자 교정에 매우 유연하게 활용됩니다.
Cas9은 indel 변이, 특정 서열 치환/삽입, 대규모 결실 또는 유전체 재배열을 유도할 수 있으며, 내인성 유전자 발현 조절이나 히스톤 변형, DNA 메틸화 조절에도 사용됩니다.
Cas9은 Streptococcus pyogenes (SpCas9) 또는 Staphylococcus aureus (SaCas9)에서 유래할 수 있습니다. SaCas9은 작은 크기, 높은 효율, 닉케이스 활성, 우수한 특이성을 지녀 유전체 편집에 유리합니다. SaCas9은 NNGRRT PAM을 인식하며, 20–24 뉴클레오타이드 길이의 스페이서를 가진 DNA를 절단합니다. SpCas9과 비교해 SaCas9은 더 작으며(1368 AA vs 1053 AA), 유사한 indel 생성률을 보여 AAV 기반 all-in-one Cas9 및 gRNA 전달에 적합합니다.
