Androgen receptor ligand binding domain (AR-LBD) is a 64.42 kDa recombinant rat protein, expressed as a GST- and His-tagged protein in insect cells. The protein includes the hinge region and an LBD amino acid sequence identical to the human LBD, with differences in the hinge region at human amino acids 646, 649, 650, 655, 656, 657, and 660.
AR-LBD is expressed and purified in the absence of exogenous ligands, making it ideal for ligand-binding and coregulator recruitment studies.

Target Information
The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and the androgen-binding domain. AR acts as a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
The AR signaling pathway is crucial for the development and function of male reproductive organs such as the prostate and epididymis, and also plays roles in muscle, hair follicles, and brain. AR is a phosphoprotein that regulates MAP kinase activity, and MEK1/2 pathway inhibition correlates with changes in AR phosphorylation state. Abnormal AR signaling is linked to diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene are associated with complete androgen insensitivity (CAIS).
