Target Information

RIPK1은 serine-threonine kinase로, death receptor 결합, 병원체 인식 수용체(PRRs) 활성화, DNA 손상 등에 의해 염증 및 세포사멸 신호를 전달합니다. TNF-alpha family cytokine에 의해 TNFR1이 활성화되면 TRADD와 TRAF2가 수용체에 결합합니다. RIPK1은 DAB2IP의 Ser-728을 인산화하여 MAP3K5-JNK 세포사멸 경로를 활성화합니다. TRAF2에 의해 Lys-63 연결 사슬 형태로 유비퀴틴화되어 MAPK 및 NF-κB 경로의 신호 전달을 강화합니다. Polyubiquitinated RIPK1은 IKBKG/NEMO와 결합하여 NF-κB 활성화를 유도하며, RIPK3와의 결합은 괴사성 세포사멸을 유도합니다.
