Cyclic adenosine monophosphate (cAMP) is a key intracellular second messenger involved in signal transduction following various extracellular stimuli.
G-Protein Coupled Receptors (GPCRs) are the largest family of cell surface receptors activated by ligands such as neurotransmitters, hormones, ions, small molecules, and peptides. Ligand binding activates G-proteins, which in turn stimulate adenylyl cyclase to produce cAMP.
cAMP activates protein kinase, leading to phosphorylation of substrate proteins. GPCRs are major drug targets, with approximately 367 identified as potential candidates, though only about 20 have been successfully developed into commercial drugs.
Due to its amplification role in signal transduction, cAMP is widely used to monitor GPCR activation in therapeutic drug discovery.
