Product Specific Information

Description:
The monoclonal antibody IL22JOP reacts with and inhibits the bioactivity of human and mouse IL-22. IL-22 is a 20 kDa member of the IL-10 cytokine family secreted mainly by Th17 cells, NK cells, and other T cells. IL-23 can induce higher IL-22 levels in Th17 cells compared to IL-6 or TGF-β, suggesting IL-22 secretion by fully differentiated Th17 cells in vivo. IL-22 plays a protective role against bacterial infections in lungs and gut. Elevated IL-22 production has been observed in asthma, ankylosing spondylitis, and rheumatoid arthritis patients.

IL22JOP is also reported to recognize rhesus IL-22.

Applications Reported:
Reacts with and inhibits the bioactivity of human and mouse IL-22.

Applications Tested:
The ND50 of IL22JOP, determined by inhibition of IL-10 induction in COLO205 cells by IL-22, is 10–40 ng/mL in the presence of 0.2 ng/mL recombinant human or mouse IL-22. Neutralization dose varies with assay method, cytokine concentration, and cell type.

Target Information

IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine belonging to the IFN-IL-10 family (including IL-19, IL-20, IL-24, IL-26, IL-28, IL-29, and interferons). It is primarily produced by activated T cells and NK cells. The IL-22 gene is located on chromosome 12q and structurally related to IL-10.

IL-22 acts via the heterodimeric receptor complex IL-22R1 and IL-10R2, activating the JAK-STAT signaling pathway. A soluble receptor, IL-22BP, antagonizes IL-22 signaling. IL-22 mainly targets nonimmune cells such as epithelial cells, hepatocytes, and keratinocytes, influencing innate immunity, inflammation, and tissue repair. It is upregulated during chronic inflammation and plays a key role in host defense and wound healing.
