Target Information

CD172a, also known as signal-regulatory protein alpha, is a receptor-type transmembrane glycoprotein expressed on cells of myeloid origin, including granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells.
It acts as a substrate for activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase-coupled signaling pathways.
Binding of CD172a to CD47 triggers phosphorylation of ITIMs, recruiting tyrosine phosphatases SHP-1 and SHP-2, which regulate cellular functions through substrate dephosphorylation.
CD172a–CD47 ligation is involved in inhibition of macrophage phagocytosis and bidirectional activation of T cells and dendritic cells.
