Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor is over 90 kDa and has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain.
AR is a ligand-activated transcription factor that binds active testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

The AR signaling pathway is essential for the development and function of male reproductive organs such as the prostate and epididymis, and also plays roles in muscle, hair follicles, and brain. AR is a phosphoprotein that regulates MAP kinase activity, and MEK1/2 pathway inhibition correlates with changes in AR phosphorylation.

Abnormalities in AR signaling are associated with diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene are linked to complete androgen insensitivity syndrome (CAIS).
