Product Specific Information

• Specificity: Recognizes an epitope on CD35 (CR1, 190 kDa), but not on CD21 (CR2, 145–150 kDa), which are alternative splice variants of primary Cr2 gene transcripts.

Target Information

CD35 (CR1, Complement Receptor 1) is a cell membrane-bound, monomeric glycoprotein primarily functioning as a receptor for C3b and C4b, key components of the complement system responsible for clearance of foreign macromolecules.
It mediates cellular binding to particles and immune complexes that have activated complement.

The CD35 complex includes four allotypes (C, A, B, D) with molecular weights of approximately 160, 190, 220, and 250 kDa, respectively.
CD35 is involved in immune complex processing, promoting binding and phagocytosis of C3b complexes, and inhibiting complement activation.

Expression:
Found on neutrophils, eosinophils, monocytes, B-cells, some NK-cells, erythrocytes, myeloid leukemias, and follicular dendritic reticulum cells; negative on basophils.

Clinical Relevance:
Reduced expression or mutations in the CD35 gene are associated with gallbladder carcinoma, mesangiocapillary glomerulonephritis, systemic lupus erythematosus, and sarcoidosis.
Certain mutations confer protection against severe malaria by reducing Plasmodium falciparum rosetting.
Alternate allele-specific splice variants encoding different isoforms have been identified, and a secreted form of CD35 has been described though not fully characterized.
