Target Information
F-Box and WD Repeat Domain Containing 7 (FBXW7) is a member of the F-box protein family characterized by an approximately 40 amino acid F-box motif.
F-box proteins form part of the SCF (SKP1-cullin-F-box) ubiquitin ligase complex, functioning in phosphorylation-dependent ubiquitination.
They are divided into three classes:
- Fbws: containing WD-40 domains
- Fbls: containing leucine-rich repeats
- Fbxs: containing other interaction modules or no recognizable motifs
FBXW7 (formerly FBX30) belongs to the Fbws class and contains 7 tandem WD40 repeats.
It binds directly to cyclin E and likely targets it for ubiquitin-mediated degradation.
Mutations in this gene have been observed in ovarian and breast cancer cell lines, suggesting its role in cancer pathogenesis.
Multiple transcript variants encoding different isoforms have been identified.
