Target Information
SMCR8는 C9orf72-SMCR8 복합체의 구성 요소로, guanine nucleotide exchange factor (GEF) 활성을 가지며 자가포식을 조절합니다. 이 복합체는 GDP 결합형 RAB8A 및 RAB39B를 GTP 결합형으로 전환시켜 자가포식소 성숙을 촉진합니다. 또한 ATG1/ULK1 kinase 복합체와 상호작용하여 단백질 인산화 활성을 억제함으로써 자가포식 개시를 음성 조절합니다. SMCR8는 mTORC1 신호를 조절하며, 세포질뿐 아니라 핵에서도 작용하여 ULK1 및 WIPI2 유전자 발현을 억제합니다.
