Target Information

SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2), also known as 2019-nCoV, causes illnesses ranging from mild cold-like symptoms to severe respiratory distress and death.
The Spike (S) protein of SARS-CoV-2 is crucial for viral attachment, fusion, and entry, making it a key target for antibody, inhibitor, and vaccine development.
The receptor-binding domain (RBD, S-RBD) in the S1 subunit binds strongly to human and bat ACE2 receptors, showing higher affinity than that of SARS-CoV.
S1 contains the RBD responsible for ACE2 binding, while S2 mediates membrane fusion.
The full-length SARS-CoV-2 S protein is 1273 amino acids long, including:

  • Signal peptide: residues 1–13
  • S1 subunit: residues 14–685
  • S2 subunit: residues 686–1273
    The S-ECD represents the extracellular domain of the Spike protein.
