Product Specific Information
The 15B8 monoclonal antibody reacts with human and mouse beta-catenin, a member of the catenin family involved in cellular adhesion and Wnt signaling. Beta-catenin binds to the cytoplasmic tail of E-cadherin and plays a crucial role in mediating cellular adhesion and gene regulation. Upon Wnt activation, beta-catenin translocates to the nucleus and interacts with transcription factors TCF and LEF to modulate gene expression.
Expression of beta-catenin occurs in both immune and non-immune tissues, including thymocytes and lymphocytes. The Wnt/beta-catenin pathway is essential for T, B, and hematopoietic stem cell development.
Applications Reported: Suitable for intracellular staining and flow cytometric analysis, immunoprecipitation, western blotting, and immunohistochemical staining of formalin-fixed paraffin-embedded tissues.
Applications Tested: Validated for western blot (Jurkat cell lysates, 1–5 µg/mL) and IHC (≤5 µg/mL, low or high pH antigen retrieval). Optimal titration recommended.
Target Information
Beta-catenin is an adherens junction protein critical for epithelial cell layer formation and intercellular adhesion. It interacts with E-cadherin and alpha-catenin, linking the adhesion complex to the actin cytoskeleton. Beta-catenin also associates with APC and microtubules, participating in multiple signaling pathways, including Wnt/Wingless.
Pathological roles of beta-catenin have been identified in pilomatrixoma, medulloblastoma, colorectal, ovarian, and other solid tumors. In the nucleus, beta-catenin co-activates Lef/Tcf transcription factors, promoting transcription of genes such as cyclin D and c-myc, which drive cell proliferation and tumorigenesis.
