Target Information
CHMP3 is a probable core component of the endosomal sorting complex required for transport (ESCRT-III), which is involved in multivesicular body (MVB) formation and sorting of endosomal cargo proteins. MVBs contain intraluminal vesicles (ILVs) generated by invagination and scission from the endosomal membrane, leading to lysosomal degradation of membrane proteins such as growth factor receptors and lipids.
The ESCRT machinery functions in membrane fission events including cytokinesis and viral budding (e.g., HIV-1). ESCRT-III proteins mediate vesicle extrusion and membrane fission, possibly with AAA ATPase VPS4. CHMP3 selectively binds phosphatidylinositol 3,5-bisphosphate (PtdIns(3,5)P2) and PtdIns(3,4)P2, and plays a role in endosomal sorting and trafficking of the EGF receptor. Isoform 2 prevents stress-mediated cell death and accumulation of reactive oxygen species in yeast cells.
