Target Information
Apolipoprotein B (apo B)는 인간 혈장에서 저밀도 지단백(LDL)의 주요 단백질로 약 260–500 kDa의 분자량을 가진다. Apo B는 세포막의 특정 수용체에 결합하여 LDL 및 VLDL 콜레스테롤을 제거하는 데 관여한다. LDL은 혈중 콜레스테롤 운반체로서 모세혈관벽의 수용체에 결합하여 콜레스테롤의 흡수와 제거를 매개한다.
동맥경화 병변에서는 산화된 LDL이 발견되며, 이는 대식세포의 scavenger receptor A 및 CD36을 통해 인식되어 섭취된다. 산화된 LDL은 동맥경화의 지표로 간주되며, 산화 형태에 따라 생화학적 특성이 다르다.
Apolipoprotein B는 주로 간에서 합성되며, VLDL, IDL, LDL 및 lipoprotein(a)의 주요 구성 요소이다. LDL 수용체의 리간드 역할을 하며, 혈중 농도 상승은 조기 동맥경화와 관련이 있다.
혈장 내 Apolipoprotein B는 apoB-48과 apoB-100 두 가지 주요 형태로 존재하며, 동일한 유전자에서 RNA 편집을 통해 생성된다. APOB 유전자 또는 조절 영역의 돌연변이는 hypobetalipoproteinemia, normotriglyceridemic hypobetalipoproteinemia, hypercholesterolemia 등 혈중 콜레스테롤 및 Apolipoprotein B 수준에 영향을 미치는 질환을 유발할 수 있다.
