Target Information
GAPDH(Glyceraldehyde-3-phosphate dehydrogenase)는 해당과정(glycolysis)에 관여하는 촉매 효소로, 동일한 37 kDa 서브유닛 4개로 구성된 테트라머 형태로 존재합니다. NADPH 존재하에 1,3-bisphosphoglycerate를 glyceraldehyde 3-phosphate로 가역적으로 환원시킵니다.
해당과정 외에도 GAPDH는 DNA 복제, DNA 수선, RNA 수출, 막 융합, 미세소관 결합 등 다양한 세포 기능에 관여합니다. 또한, 세포사멸(apoptosis)과 신경퇴행성 질환의 유전자 발현 조절에도 관여하는 것으로 알려져 있습니다. GAPDH는 Alzheimer’s disease 관련 단백질(APP) 및 Huntington’s disease 관련 단백질(Huntingtin)과 상호작용하며, actin 및 tubulin과의 결합도 보고되었습니다.
GAPDH는 대부분의 조직에서 높은 수준으로 발현되므로 Western blot 분석 시 단백질 적재 대조군(loading control)으로 유용하게 사용됩니다.
