Target Information
Smad4는 인간의 18번 염색체에 위치한 Smad4 유전자에 의해 암호화됩니다.
Smad4는 Smad family member 4, Sma- and Mad-related protein 4 또는 Mothers against decapentaplegic (MAD) homolog 4로도 알려져 있습니다.
이 단백질은 N-말단의 MH1 (MAD homology 1) 도메인과 C-말단의 MH2 (MAD homology 2) 도메인을 가지며, 각각 DNA 결합과 단백질 상호작용에 관여합니다.
TGF-beta 계열 리간드(Transforming Growth Factor-beta, BMP 등)가 수용체에 결합하면 Smad1, 2, 3, 5, 8 (R-Smad)이 인산화되어 Smad4 (co-Smad)와 삼량체를 형성하고, 이 복합체가 핵으로 이동합니다.
Smad6, 7 등의 억제성 Smad (I-Smad)는 활성화된 R-Smad와 상호작용하여 신호 경로를 억제합니다.
Smad4는 Cyclin D1의 하향 조절 및 콜라겐의 상향 조절을 통해 세포 증식을 억제하는 종양 억제 단백질로 작용합니다.
탈인산화 과정은 Smad 단백질의 핵 내외 이동을 조절합니다.
