Target Information
Smad4는 인간 18번 염색체에 위치한 Smad4 유전자에 의해 암호화되며, Smad family member 4, MAD homolog 4 등으로도 알려져 있습니다.
Smad4는 N-말단의 MH1 (MAD homology 1) 도메인과 C-말단의 MH2 (MAD homology 2) 도메인을 가지며, 각각 DNA 결합 및 단백질 상호작용에 관여합니다.
TGF-β 및 BMP 등의 리간드가 수용체에 결합하면 R-Smad (Smad1, 2, 3, 5, 8)가 인산화되어 Smad4와 삼합체를 형성하고 핵으로 이동합니다.
Smad4는 Cyclin D1 억제, Collagen 발현 증가 등 표적 유전자 전사를 조절하여 종양 억제 단백질로 작용합니다.
Smad6 및 Smad7과 같은 억제성 Smad는 활성화된 R-Smad와 상호작용하여 신호 전달을 조절합니다.
탈인산화 과정은 Smad의 핵 내외 이동 및 동역학을 조절합니다.
