Product Specific Information

• Reconstitute with 1 mL of distilled water.
• After reconstitution, store undiluted at 4°C in the dark.
• For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
• Mouse anti-Rat CD161 antibody (clone 10/78) recognizes the rat Killer cell lectin-like receptor subfamily B protein (NKR-PI or CD161).

Target Information

KLRB1 (Killer cell lectin-like receptor subfamily B, member 1; CD161) is a type II membrane protein with an external C terminus. It is expressed on nearly all NK cells and a subset of CD3⁺ T cells.
KLRB1 forms a homodimeric surface protein (40–44 kDa) and functions as an inhibitory receptor on NK cells. Upon activation, it stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide, activating AKT1/PKB and RPS6KA1/RSK1 kinases, promoting T cell proliferation.
KLRB1 binds carbohydrate epitopes such as Gal-α(1,3)Gal and N-acetyllactosamine, and interacts with CLEC2D/LLT1 to inhibit NK cell cytotoxicity and interferon-gamma secretion. The protein includes extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
