Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
Cyclin E belongs to the highly conserved cyclin family, characterized by periodic changes in protein abundance through the cell cycle. Cyclins regulate CDK kinases and exhibit distinct expression and degradation patterns, coordinating mitotic events temporally. Cyclin E forms a complex with CDK2, essential for the G1/S transition. It accumulates at the G1-S phase boundary and is degraded during S phase. Overexpression has been observed in many tumors, contributing to chromosomal instability and tumorigenesis. Cyclin E also associates with NPAT, involved in histone gene expression and cell-cycle progression without pRB. Two alternatively spliced transcript variants encoding distinct isoforms have been described; additional variants exist but are not fully characterized.
