Product Specific Information

Specificity: Recognizes a 48 kD Type I glycoprotein belonging to the TNFR superfamily. Expressed on normal or malignant B cells, macrophages, follicular dendritic cells, endothelial cells, fibroblasts, and at low levels on plasma cells.


Target Information

CD40 is a single-chain glycoprotein and a member of the tumor necrosis factor/nerve growth factor superfamily. It is expressed on all B cells except plasma cells, and also found on epithelial cells, carcinomas, and lymphoid dendritic cells.

CD40 plays a crucial role in immune and inflammatory responses, including:

  • T cell-dependent immunoglobulin class switching
  • Memory B cell development
  • Germinal center formation

The AT-hook transcription factor AKNA regulates CD40 expression, influencing homotypic cell interactions. TNFR2 adaptor protein interacts with CD40 to mediate signal transduction.

CD40-CD40L interaction is essential for amyloid-beta-induced microglial activation, suggesting an early role in Alzheimer’s disease pathogenesis.

Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
