Specificity: Human CD34, an approximately 105–120 kDa type I transmembrane glycoprotein expressed on the surface of most human hematopoietic stem and progenitor cells (HSPCs), as well as on mesenchymal stem cells, embryonic fibroblasts, endothelial cells, neurons, and some tumor cell lines.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) present on many stem cell populations. It serves as a marker for hematopoietic stem cells, though its expression is reversible. CD34 may function as a receptor involved in endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic and progenitor cells.

It likely mediates attachment of stem cells to the bone marrow extracellular matrix or stromal cells and may act as a scaffold for binding lineage-specific glycans, enabling interactions with lectins on stromal or marrow components. CD34 is also implicated in presenting carbohydrate ligands to selectins. Its intracellular domain is a phosphorylation site for protein kinase C, suggesting a role in signal transduction.

Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
