This peptide corresponds to a 19 amino acid peptide near the center of human SHANK3.
PEP-1426 can be used as a blocking peptide with polyclonal antibody PA5-34383.

Target Information

SHANK3 is a major scaffold postsynaptic density protein that interacts with multiple proteins and complexes to orchestrate dendritic spine and synapse formation, maturation, and maintenance.
It connects receptors of the postsynaptic membrane, including NMDA-type and metabotropic glutamate receptors, via complexes with GKAP/PSD-95 and HOMER, and the actin-based cytoskeleton.
SHANK3 plays a role in structural and functional organization of dendritic spine and synaptic junction through interactions with Arp2/3 and WAVE1 complexes, promoting F-actin clusters.
By controlling actin dynamics, SHANK3 regulates developing neuron growth cone motility and NMDA receptor signaling. It also modulates GRIA1 exocytosis and GRM5/MGLUR5 expression to control AMPA and metabotropic glutamate receptor-mediated synaptic transmission and plasticity.
SHANK3 may be required at early stages of synapse formation and can be inhibited by IGF1 to promote synapse maturation. (Source: Uniprot)
