Product Specific Information

This F38-2E2 monoclonal antibody reacts with human CD366 (TIM3 or HAVCR2). CD366 is expressed on activated CD4+ T cell subsets (Th1, Th17, Treg), CD8+ T cells, monocytes, dendritic cells, and mast cells. It exists as both membrane-bound and soluble forms, with Galectin-9 identified as its ligand. CD366 negatively regulates CD4+ T cells and influences Th1/Th17 cytokine secretion, playing roles in tolerance, autoimmune diseases, and HIV infection.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on stimulated human peripheral blood cells (5 µL/test, 100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is compatible with instruments equipped with a 355 nm ultraviolet laser.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401) or Brilliant Stain Buffer (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark.
BUV805 is compatible with short-term methanol fixation but should not be stored in methanol-containing buffers for longer than one hour.

Target Information

TIM3 (HAVCR2) is a 281-amino acid, type I cell surface glycoprotein expressed on terminally differentiated CD4+ Th1 and CD8+ Tc1 cells. It contains an IgV-like domain, a mucin-like domain, and a cytoplasmic tyrosine phosphorylation motif. TIM3 modulates macrophage activation, autoimmune disease induction, and down-regulates Th1-mediated immune responses, promoting immune tolerance. It is associated with experimental autoimmune encephalomyelitis (EAE) and other Th1-dependent autoimmune conditions.
