Mouse anti-Human CD161 antibody, clone B199.2, recognizes the human Killer cell lectin-like receptor subfamily B member 1 (KLRB1), also known as CD161, C-type lectin domain family 5 member B, HNKR-P1a, NKR-P1A, or Natural killer cell surface protein P1A.

Target Information

KLRB1 (Killer cell lectin-like receptor subfamily B, member 1, CD161) is a type II membrane protein with an external C terminus. It is expressed on most NK cells and a subset of CD3⁺ T cells. KLRB1 is a homodimeric surface protein (~40–44 kDa) that plays an inhibitory role in NK cell function.

Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide, leading to activation of AKT1/PKB and RPS6KA1/RSK1 kinases, and promoting T cell proliferation via anti-CD3 stimulation. KLRB1 acts as a lectin binding to Gal-α(1,3)Gal and N-acetyllactosamine epitopes, and also binds CLEC2D/LLT1, inhibiting NK cell-mediated cytotoxicity and interferon-γ secretion.

The protein includes extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
