PA1-86558 specifically recognizes the 180 kDa NR2B subunit of the NMDA receptor when phosphorylated at Tyr1472.
Predicted to react with bovine, chicken, dog, human, monkey, mouse, and zebrafish based on sequence homology.
- For detection of phosphoproteins, add 1 mM sodium orthovanadate to the sample buffer.
- Avoid casein-based blocking solutions (e.g., milk) as casein is a phosphoprotein and may cause high background.
- In Western blot, detects bands at approximately 180 kDa, 65 kDa, and 115 kDa in rat brain hippocampal lysate.
Target Information
NMDAR2B is a member of the N-methyl-D-aspartate (NMDA) class of ionotropic glutamate receptors.
NMDA receptors are involved in long-term potentiation, a mechanism underlying memory and learning.
They play a role in neuronal development and are implicated in disorders such as Alzheimer’s disease, epilepsy, and ischemic neuronal cell death.
Overexpression of NR2B is linked to enhanced learning and memory, while reduced expression is observed in aged or memory-impaired animals.
Phosphorylation at Tyr1472 may increase synaptic expression of NMDAR, contributing to synaptic plasticity.
NMDAR2B forms a heterotrimer with NR1, NR2, and NR3 subunits.
Diseases associated with NMDAR2B dysfunction include epileptic encephalopathy and autosomal dominant mental retardation type 6.
