Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven membrane-spanning domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also mediates HIV-2 binding and fusion through envelope glycoprotein gp120. CXCR4 is the receptor for the chemokine CXCL12/SDF-1, leading to intracellular calcium increase and MAPK1/MAPK3 activation. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP levels.

CXCR4 plays roles in hematopoiesis, cardiac ventricular septum formation, and vascularization of the gastrointestinal tract by regulating endothelial cell remodeling. In the CNS, it contributes to hippocampal neuron survival and cerebellar development. CXCR4 binds bacterial LPS and mediates inflammatory responses such as TNF secretion by monocytes. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection of human target cells. The amino-terminal domain and second extracellular loop of CXCR4 serve as HIV binding sites.
