Target Information
ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1, is a human homologue of the yeast Ire1 gene product.
It possesses intrinsic kinase and endoribonuclease activities and plays a key role in endoplasmic reticulum-based stress signaling.
ERN1 regulates IRE1 proteolysis in mammalian cells and is critical for fetal viability.
IRE1 acts as a sensor of unfolded proteins in the ER, initiating transmittance of the unfolded protein signal to the nucleus by splicing XBP1 mRNA, which converts it into a potent transcriptional activator.
Null mutants are viable under normal conditions but essential under stress conditions that cause unfolded protein accumulation in the ER.
Depletion of IRE1p through expression of a dominant negative form does not affect transfected cells under normal conditions but leads to apoptosis under stress.
