Product Specific Information

The CBRM1/5 monoclonal antibody reacts with an activation-specific epitope of human Mac-1. It binds a subset of Mac-1 molecules on neutrophils and monocytes after stimulation with chemoattractants or phorbol esters, but not on resting myeloid cells. Mac-1 participates in adhesive cell interactions, and this antibody recognizes an epitope localized to the I domain on the alpha chain near the ligand binding site.
CBRM1/5 blocks Mac-1-dependent adhesion to fibrinogen and ICAM-1 and inhibits chemoattractant-stimulated adhesion of eosinophils to ICAM-1.
Low-level activation may occur during processing of freshly drawn blood, showing some binding in unstimulated samples. Higher Mac-1 expression is observed in activated samples compared to resting cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry on stimulated peripheral blood cells.
Use at 5 µL (0.5 µg) per test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that associates with integrin beta-2 (CD18). The CD11b/CD18 complex (Mac-1, CR3) is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cell subsets, dendritic cells, and microglia.
It functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X — mediating critical cell-cell and cell-matrix interactions.
CD11b expression varies across tissues: ~8% of spleen cells, 44% of bone marrow cells, <1% of thymocytes. Commonly used as a microglial marker.
Expression increases during monocyte maturation; peritoneal macrophages express higher levels than splenic macrophages.
Dysfunction of CD11b is associated with systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune diseases.
