Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, mediating inhibitory signaling and receptor internalization via ubiquitin pathways.
CD33 is expressed on cells of the myelomonocytic lineage, including granulocyte and macrophage precursors in the bone marrow, but not on pluripotent stem cells. It binds sialic acid residues on N- and O-glycans and is a key therapeutic and diagnostic target in acute myeloid leukemia (AML).
Additionally, CD33 serves as a useful marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias. Dysregulation of CD33 is associated with diseases such as gallbladder lymphoma and extracutaneous mastocytoma.
