Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and are involved in ubiquitin-mediated receptor removal from the cell surface.
CD33 is expressed on cells of the myelomonocytic lineage, binding sialic acid residues in N- and O-glycans on cell surfaces. It serves as a therapeutic target for acute myeloid leukemia (AML).
Additionally, CD33 is present on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells, and is expressed on peripheral monocytes. It is a useful marker to distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
