Target Information

CD40 antigen is a single-chain glycoprotein belonging to the tumor necrosis factor/nerve growth factor superfamily. It shares significant homology with CD30, associated with Hodgkin’s disease. CD40 is expressed on all B cells except plasma cells, as well as on some epithelial cells, carcinomas, and lymphoid dendritic cells.
It plays an essential role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
The AT-hook transcription factor AKNA regulates CD40 expression, facilitating homotypic cell interactions. TNFR2 adaptor protein interacts with CD40 to mediate signal transduction.
CD40-CD40L interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
