Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating the cell surface expression of the TCR complex.
T cell activation through the TCR involves the cytoplasmic tails of CD3γ, CD3δ, CD3ε, and CD3ζ subunits. These are structurally related members of the immunoglobulin superfamily encoded by closely linked genes on human chromosome 11.

CD3 components have long cytoplasmic tails that associate with cytoplasmic signal transduction molecules via a double tyrosine-based motif. CD3 may play roles in TCR-induced growth arrest, survival, and proliferation.

The CD3 antigen is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Decreased T lymphocyte percentages may be observed in some autoimmune diseases.

Genes encoding CD3ε, CD3γ, and CD3δ are located on chromosome 11. Defects in CD3 genes are associated with CD3 immunodeficiency.
