Target Information
The MYH9 gene, located on chromosome 22q12.3, encodes the heavy chain of non-muscle myosin IIA (NMHC IIA), a crucial actin cytoskeleton component involved in cell migration, adhesion, division, and shape maintenance.
The gene spans over 106 kb with 41 exons encoding a 1,960 amino acid protein forming a hexameric complex (two heavy chains, two regulatory light chains, two essential light chains).
Mutations in MYH9 cause autosomal dominant MYH9-related diseases (MYH9-RD), such as May-Hegglin anomaly, Fechtner syndrome, and Epstein syndrome, characterized by macrothrombocytopenia and potential hearing loss, renal failure, and cataracts.
MYH9 is also vital in hematopoiesis, maintaining hematopoietic stem and progenitor cell survival; its dysfunction leads to blood cell deficiencies and bone marrow failure.
