Product Specific Information

The M18/2 monoclonal antibody reacts with mouse CD18, a 90–95 kDa integrin beta 2 glycoprotein. CD18 is the common beta subunit that associates with CD11a (alpha L), CD11b (alpha M), and CD11c (alpha X) to form LFA-1, Mac-1, and gp150/95 complexes. These heterodimers are expressed on all leukocytes, with NK and T cells showing higher surface density. Through interactions with their counter-receptors, CD18 complexes are involved in cellular adhesion and immune responses.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes and bone marrow cells; recommended ≤1 µg per test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information

CD18 (integrin beta 2 subunit) is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11a–d to constitute leukocyte (β2) integrins:

• αLβ2 (CD11a/CD18, LFA-1)
• αMβ2 (CD11b/CD18, Mac-1, CR3)
• αXβ2 (CD11c/CD18)
• αDβ2 (CD11d/CD18)

These integrins mediate cell adhesion and signaling essential for leukocyte migration and immune function. Deficiency in CD18 leads to leukocyte adhesion deficiency type I (LAD1), and reduced expression is associated with psoriasiform skin disease. CD18 also mediates leukotoxin-induced cytolysis by Mannheimia haemolytica.
