GRAMD4 (GRAM domain-containing protein 4) maps to human chromosome 22, which houses over 500 genes. Also known as death-inducing protein (DIP), GRAMD4 is a 578 amino acid mitochondrial membrane protein involved in the p53-independent E2F-1 death pathway, often deregulated in various cancers. Overexpression triggers apoptosis via caspase-3 activation and PARP cleavage. GRAMD4 is expressed in lung tissue and lung squamous cell carcinoma (LSCC), with upregulation by E2F1 after 4-hydroxytamoxifen treatment.

Target Information
GRAMD4, also known as Death Inducing Protein, is a 578 amino acid protein containing one GRAM domain. It is a multi-pass membrane protein localized to the mitochondrial membrane. Overexpression induces apoptosis through caspase-3 activation and PARP cleavage. GRAMD4 is expressed in lung tissue and primary LSCC, showing mitochondrial upregulation by E2F-1 after 4-hydroxytamoxifen exposure. These findings suggest GRAMD4 as a potential cancer therapy target.
