Target Information
DOK6 (Docking Protein 6)은 p62 Dok 패밀리의 Dok-4/5 subclass에 속하는 새로운 세포내 어댑터 단백질입니다.
이 단백질은 발달 중인 중추신경계에서 높게 발현되며, Ret 수용체와 함께 교감, 감각, 부교감 신경절 및 신장 요관 싹(ureteric buds) 등에서 공발현됩니다.
Pull-down assay 결과, Dok-6의 phosphotyrosine binding (PTB) 도메인은 GDNF-활성화 Ret의 인산화된 Tyr(1062) 잔기와 결합함을 보여줍니다.
또한, Ret의 리간드 활성화는 Src 의존적 메커니즘을 통해 Dok-6 C-말단의 티로신 잔기를 인산화시키며, 이는 Dok-6이 Ret-Src 신호전달 경로의 기질임을 시사합니다.
