Target Information
MAP4K1 (also known as HPK1) or Hematopoietic progenitor kinase 1 is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily. It acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway.
MAP4K1 reduces T cell receptor (TCR) signaling activity and T cell proliferation by phosphorylating the adaptor protein SLP-76.
JNKs/SAPKs play a crucial role in stress responses in mammalian cells. Understanding MAP4K1 function is key to elucidating the molecular basis of blood cell differentiation.
HPK1 is predominantly expressed in hematopoietic progenitor cells and activates JNK1 specifically, enhancing AP-1-mediated transcriptional activity. HPK1 binds and phosphorylates MEKK1 directly, and JNK1 activation by HPK1 is inhibited by dominant-negative MEKK1 or MKK4/SEK mutants.
Unlike PAK65, HPK1 does not contain a Rac1/Cdc42-binding domain and does not interact with these small GTPases.
