Target Information

Human Rad18 (hRad18) is a homologue of S. pombe Rad18. In yeast, Rad6 and Rad18 play crucial roles in post-replication repair by filling gaps in daughter strands during replication of damaged DNA. The Rad6–Rad18 interaction is critical at early steps of lesion bypass. Rad18 forms a tight complex with Rad6 via a conserved ring-finger motif, allowing binding to damaged DNA through single-stranded DNA affinity. This complex facilitates degradation of stalled DNA replication components via the ubiquitin-conjugating activity of Rad6.
Human RAD18 maps to chromosome 3p24–25, a region frequently deleted in lung, breast, ovary, and testis cancers. The RAD18 gene encodes a 495-amino-acid protein (~56 kDa) and is ubiquitously expressed in human tissues.
