Target Information

PYCARD functions as a key mediator in apoptosis and inflammation. It promotes caspase-mediated apoptosis involving caspase-8 and -9, and is involved in activation of the mitochondrial apoptotic pathway. PYCARD mediates mitochondrial translocation of BAX and activates BAX-dependent apoptosis coupled with activation of caspase-9, -2, and -3.

It plays a central role in macrophage pyroptosis, forming the ASC pyroptosome that recruits and activates caspase-1. As an inflammasome adapter, PYCARD mediates activation of caspase-1 leading to processing and secretion of proinflammatory cytokines. It interacts with pattern recognition receptors such as NLRP2, NLRP3, AIM2, and possibly IFI16.

Additionally, it is involved in dendritic cell maturation, T-cell activation, cytoskeletal rearrangements, and transcriptional activation of cytokines and chemokines via AP-1, NF-κB, and MAPK signaling pathways. Isoforms 2 and 3 may modulate inflammasome activity differently.
