Target Information
The protein tyrosine phosphatase PTPN22 (PTP22, LYP, PEP, formerly PTPN8) is a genetic variant associated with increased risk of autoimmune diseases such as type 1 diabetes and rheumatoid arthritis.
The minor allele of a missense SNP in PTPN22 encodes a hematopoietic-specific phosphatase known as Lyp, found in ~17% of the general Caucasian population and ~28% of those with rheumatoid arthritis.
This allele may disrupt the P1 proline-rich motif crucial for interaction with the SH3 domain of CSK, potentially affecting normal T-cell regulation.
PTPN22 is considered an effector and/or regulator of CSK in T cells and other hematopoietic cells.
